I have got a web template but I want to customize it using joomla. Is it possible. Please let me know.

Joomla is a content management system, not a template editor.

It *uses* one or more templates in order to display content stored in a database to users, but it is not designed to customize them.

That being said, some Joomla Templates can be "tweaked" via parameters that can be tuned from the Joomla administrative backend.

A template editing capability could also be added through Joomla extensions, but I’m not personally aware of any such extensions at this time.

Only if it is a Joomla Template. If it is and you know some HTML you can go into the template are and modify the CSS and HTML. You can also do this by downloading the index.php and Styles.css files and edit them in a text editor, or dreamweaver or something like that.
